Check your firewall or AV programs arent blocking lotro.exe, a lot of us get this issue after nearly major every update. Heuristic scans are what usually block it on my Comodo firewall; whitelisting doesnt always work, and you have to re-whitelist it several times before the firewall will stop blocking it.
